چکیده
N-glycosylation is a post-translational modification of proteins that occurs across all three domains life. In Archaea, crucial for cell stability and motility, but importantly also has significant implications virus–host interactions. While some archaeal viruses present glycosylated or interact with host proteins, the direct influence on interactions remains to be elucidated. this study, we generated an N-glycosylation-deficient mutant Halorubrum lacusprofundi, halophilic archaeon commonly used study cold adaptation, examined impact compromised infection dynamics two very diverse viruses. had no life cycle head-tailed virus HRTV-DL1, observed effect membrane-containing HFPV-1. Both intracellular genome numbers extracellular particle HFPV-1 were increased in strain, which attribute instability surface-layer builds protein envelope cell. When testing plasmid vesicles, specialized membrane vesicles transfer between cells, determined vesicle strongly dependent glycosylation machinery. Our thus provides important insight into role while pointing how differs amongst various virus-like elements.

Amino Acid Substitutions in Cold-Adapted Proteins from Halorubrum lacusprofundi, an Extremely Halophilic Microbe from Antarctica
The halophilic Archaeon Halorubrum lacusprofundi, isolated from the perennially cold and hypersaline Deep Lake in Antarctica, was recently sequenced and compared to 12 Haloarchaea from temperate climates by comparative genomics. Amino acid substitutions for 604 H. lacusprofundi proteins belonging to conserved haloarchaeal orthologous groups (cHOGs) were determined and found to occur at 7.85% of...
متن کاملMorphological and proteomic analysis of biofilms from the Antarctic archaeon, Halorubrum lacusprofundi
Biofilms enhance rates of gene exchange, access to specific nutrients, and cell survivability. Haloarchaea in Deep Lake, Antarctica, are characterized by high rates of intergenera gene exchange, metabolic specialization that promotes niche adaptation, and are exposed to high levels of UV-irradiation in summer. Halorubrum lacusprofundi from Deep Lake has previously been reported to form biofilms...
متن کاملCloning, overexpression, purification, and characterization of a polyextremophilic β-galactosidase from the Antarctic haloarchaeon Halorubrum lacusprofundi
BACKGROUND Halorubrum lacusprofundi is a cold-adapted halophilic archaeon isolated from Deep Lake, a perennially cold and hypersaline lake in Antarctica. Its genome sequencing project was recently completed, providing access to many genes predicted to encode polyextremophilic enzymes active in both extremely high salinity and cold temperatures. RESULTS Analysis of the genome sequence of H. la...
متن کاملComplete genome sequence of the Antarctic Halorubrum lacusprofundi type strain ACAM 34
Halorubrum lacusprofundi is an extreme halophile within the archaeal phylum Euryarchaeota. The type strain ACAM 34 was isolated from Deep Lake, Antarctica. H. lacusprofundi is of phylogenetic interest because it is distantly related to the haloarchaea that have previously been sequenced. It is also of interest because of its psychrotolerance. We report here the complete genome sequence of H. la...
متن کاملDeveloping a genetic manipulation system for the Antarctic archaeon, Halorubrum lacusprofundi: investigating acetamidase gene function
No systems have been reported for genetic manipulation of cold-adapted Archaea. Halorubrum lacusprofundi is an important member of Deep Lake, Antarctica (~10% of the population), and is amendable to laboratory cultivation. Here we report the development of a shuttle-vector and targeted gene-knockout system for this species. To investigate the function of acetamidase/formamidase genes, a class o...
